Our laboratory is carrying out large-scale CRISPR screens, in which we measure the effect on cell fitness when different cancer-related genes are disrupted in combination. Our approach, based on CRISPR/Cas12a, allows us to quantify the effect of perturbing more than 20,000 different gene combinations in parallel within a single screening experiment. The data from these powerful screens reveal gene targets which are synergistic with one another in particular genetic and mutational contexts, with the potential to uncover highly specific combinatorial drug treatment strategies.

This co-op project is suitable for students in their 3rd or 4th year. Previous cell culture experience is not a prerequisite, though it would allow the student to carry out screening work at an earlier stage in the co-op term.

What you’ll do:

As a co-op student in our laboratory, you will gain hands-on experience in cutting-edge CRISPR screening techniques used in cancer research. You will be involved in a number of combinatorial screens in cancer cell lines, learning and carrying out key experimental work, as well as data analysis. Responsibilities include:

  • Pre-screen characterization of cell lines – quantifying growth rates in culture, sensitivity to selection drugs (puromycin)
  • Combinatorial screening in cancer cell lines – large scale mammalian cell line culture, involving propagation of ~108 CRISPR-library-transduced cells for 4-5 weeks; extraction of DNA from cell harvests at various timepoints
  • Data analysis – analysis of large combinatorial datasets using Bayesian models, formulating hypotheses, planning follow-up experiments to validate findings

By the end of the rotation, students will:

  • Demonstrate a deep understanding of cancer functional genomics and combinatorial targeting
  • Develop expertise in cellular and molecular experimental techniques commonly used in cancer research
  • Generate high-quality data that meets standards for inclusion in a scientific publication
  • Critically analyze experimental results, identifying potential pitfalls, proposing alternative interpretations, and designing control experiments
  • Formulate and test hypotheses using data-driven insights and literature review, designing follow-up experiments to refine findings
  • Present findings effectively through clear data visualization and logical scientific communication.

Core Competencies:

  • Brings an understanding of the Indigenous specific racism and the broader systemic racism that exists in the colonial health care structure, and has demonstrated initiatives in breaking down barriers and ensuring a safe environment ensuring a sense of belonging to all and informed by Indigenous Cultural Safety.
  • Awareness of social, economic, political and historical realities of settler colonialism on Indigenous Peoples and familiarity with addressing Indigenous-specific anti-racism, anti-racism and Indigenous Cultural Safety and foundational documents and legislative commitments (the Declaration Act, the Declaration Action Plan, TRC, IPS, Remembering Keegan, etc.).

You have:

  • Demonstrates a commitment to beginning and/or continuing their personal learning journey related to Indigenous-specific racism and dismantling systems of oppression, as well as addressing racism more broadly. Shows willingness to articulate and share their learning experiences to contribute to a culture of motivation and inspiration among peers.
  • Demonstrates foundational knowledge of the social, economic, and political realities of settler-colonialism and its impacts on Indigenous peoples and equity-deserving groups within social and health contexts. Understands the impact of social determinants of health-on-health outcomes. Shows a commitment to learning about and upholding legislative obligations and provincial commitments outlined in foundational documents such as the Truth & Reconciliation Commission’s Calls to Action (2015), In Plain Sight (2020), BC's Declaration on the Rights of Indigenous Peoples Act (2019), United Nations Declaration on the Rights of Indigenous Peoples (UNDRIP), Reclaiming Power and Place: Missing and Murdered Indigenous Women & Girls Calls for Justice (2019), the Declaration Act Action Plan, Remembering Keegan: A First Nations Case Study, the BC Human Rights Code, Anti-Racism Data Act, and the Distinctions Based Approach.

Job Type: Casual (8 months)

Wage: Starting wage for this position would be determined with consideration of the successful candidate’s relevant education and experience and would be in alignment with the provincial compensation reference plan.
